| Summary, etc. | SUSPENSIOcNul tures of plant cells are becoming increasingly important as experimental material for investigations concerning plant growth and metabolism. Some of the problems and potentials of the suspension culture technique have been discussed by Street et al. [14], Carew and Staba [l], and Tulecke et al. [16]. Initial attempts to culture plant cells in liquid cultures necessitated the addition of coconut milk [ 131 or other organic supplement to the medium [15], but several cultures have now been observed to grow in chemically defined media of varying complexity. Eriksson [3]and Filner [4]have achieved good growth of cultures in simple media consisting of mineral salts, sucrose, B-vitamins, and ,4-dichlorophenoxyacetic acid (2,4-D). The simple media are useful for practical reasons, and also make the cultures more suitable for studies of metabolism and growth. Filner [5]has used a strain of tobacco cells which grows well in a medium devoid of organic nitrogen to study the repression and derepression of nitrate reductase by amino acids, and Olson [la]used the same strain of tobacco in a study of cell wall metabolism. Halperin and Wetherell [8]have studied morphogenesis in carrot cells cultured in a similar medium in which nitrogen was supplied as ammonium and nitrate and observed embryogenesis. The ammonium ions were required to ensure a consistently high percentage of embryogenesis. The present report deals with a study of some of the nutrient requirements of suspension cultures of soybean grown in a mineral salt and sucrose medium. |
