Uno de los modelos biosintéticos más estudiados en los laboratorios del mundo ha sido Catharanthus roseus, planta económicamente importante debido al valor medicinal de los alcaloides indólicos que produce. De los cultivos de raíces transformadas de C. roseus, se han reportado los alcaloides indólicos en concentraciones mayores a las obtenidas en otros tipos de cultivos in vitro. El presente estudio tuvo como objetivo la caracterización de los alcaloides de la línea J1 de raíces transformadas de C. roseus. Para este fin el extracto metanólico de las raíces fue sometido a una separación ácido-base para obtener un extracto de alcaloides totales. El trabajo de purificación se inició con una partición con disolventes de polaridad ascendente (Hx, CH2C12, AcOEt y BuOH). La fracción hexánica (AJ-6A), así como la diclorometánica (AJ-6B), se purificaron por CC "flash", CCP y CLV, obteniendo en total cinco fracciones AJ-10B, AJ-12C, AJ-15B, AJ-M12C y AJ-M18. Las dos últimas se identificaron por sus datos espectroscópicos como los alcaloides indólicos ajmalicina y epimisilina, respectivamente. |
