El presente trabajo presenta el diseño de la metodología para el aislamiento y purificación de los alcaloides no polares presentes en el cultivo de callos fotomixotróficos de Catharanthus roseus, para realizar lo anterior, se llevó a cabo una extracción ácido-base para obtener los alcaloides totales; el proceso de purificación se inició mediante la partición por polaridad de dicho extracto de alcaloides totales, utilizando cuatro diferentes disolvente, de polaridad creciente (hexano, diclorometano, acetato de etilo y butano). Posteriormente se llevó a cabo la purificación de las bandas mayoritarias de alcaloides presentes en las fracciones no polares (hexánica y diclorometánica) utilizando diferentes técnicas cromatográficas. Finalmente, nos avocamos a identificar a aquellos alcaloides que se purificaron de dichas fracciones no polares, mediante el uso de técnicas espectrosocópicas como son: resonancia magnética nuclear protónica, infrarrojo y espectrometría de masas; lograndose identificar a los siguientes alcaloides: ajmalicina, catarantina y vincamina |
