En el presente trabajo se presenta el estudio sistemático de la enzima glutamino sintetasa en hojas de Catharanthus roseus, que incluye su purificación, caracterización y el estudio de su regulación durante diferentes condiciones medio ambientales. Se determinó que la glutamino sintetasa se encuentra como dos isoenzimas en la hoja. Estas isoenzimas se denominaron GS1 y GS2. La GS1 está localizada en el citoplasma, mientras que la GS2 se encuentra dentro del cloroplasto. La masa molecular de las isoenzimas es de 46 y 48 kOa, respectivamente. Las Km para los sustratos glutamato, hidroxilamina y ATP son diferentes. La actividad de la GS2 muestra una dependencia total de la presencia de agentes reductores. La GS1 presenta mayor resistencia a la incubación a altas temperaturas que la GS2, la cual pierde totalmente su actividad después de 15 minutos a 50°C. Se demostró que la isoenzima cloroplástica está glucosilada usando tres técnicas diferentes. La presencia de nitrato en el medio provdca un aumento en la actividad de la GS solamente observado en raíces. Durante los períodos de estrés, la actividad enzimática disminuye. No hay síntesis de novo de esta enzima y hay una pérdida preferencial de la isoenzima cloroplástica. Esta pérdida se atribuye a los cambios que sufre su nivel de glucosilación. |
