Summary, etc. Byrsonima crassifolia is a Brazilian fruit, native to the Amazon biome, rich in antioxidant compounds that can be applied in the treatment of different diseases, such as cancer. However, only a few studies carry out the extraction of phenolic compounds to evaluate the cytotoxicity of compounds from this fruit in the treatment of different diseases. Thus, this work aimed to investigate the influence of the drying method (oven drying or freeze drying)on the extraction of phenolic compounds through ultrasound-assisted extraction and evaluate the cytotoxicity of the Byrsonima crassifolia fruit extracts obtained. Different temperatures, solid-to-liquid ratios, and ethanol concentrations were evaluated according to a rotational central composite design. Cell viability of CCD 1059 Sk and Sk-Mel-28 cells was evaluated using different extract concentrations (0.25 to 5 mg/mL)for 24 h of treatment. The best results in terms of total phenolic compounds (39.84 ± 0.31 mg GAE/g dry fruit)and antioxidant activity (229.31 ± 2.58 ?mol TE/g dry fruit)were for freeze-dried fruits, using a temperature, solid-to-liquid ratio, and ethanol concentration of 80 °C, 25 mg/mL, and 80 percent (v/m), respectively. Regarding cell viability, the Byrsonima crassifolia extract demonstrated positive results in terms of cell viability for healthy human cells (CCD 1059 Sk), but not inhibited Sk-Mel-28 cells. This study demonstrates that the phenolic compounds present in Byrsonima crassifolia extract have promising potential for future therapeutic applications, due their antioxidant and anti-inflammatory properties and their non-cytotoxicity in human cells.
