Summary, etc. The article introduces the concept of explant state as one of the main factors infuencing the initiation of Pinus sylvestris L. somatic embryogenesis. Data on Scots pine zygotic embryo development in trees growing on the territory of South Karelia are presented. Plant material was collected from trees of diferent age: Petrozavodsk Seed Orchard of the frst order (40 years), progeny trial in the Man’ga village (10 years), and the Petrozavodsk Park (20 years). It was found that fertilization and the subsequent formation of zygotic embryos occurred faster in trees growing in the park. For SE initiation, cleavage polyembryony appeared to be the most suitable stage of zygotic embryo development. Less frequently embryogenic tissue was formed at the later stages, i.e., globular and precotyledonary. The in vitro cultivation of 1022 megagametophytes containing immature embryos on DCR medium showed that explants collected from plus tree clones in Petrozavodsk Seed Orchard extruded (8.06±1.33percent) and formed cell mass (0.65±0.32percent) more actively than younger trees from the park, where extrusion was absent and cell mass was formed in 0.46±0.46percent of cases, as well as from progeny trial explants, where embryonic mass extrusion (0.96±0.67percent) was observed, but cell mass formation did not occur. It was recorded that only 57.5percent of cultured explants contained a zygotic embryo within the megagametophyte. This phenomenon distinctive for conifers is a result of a mechanism to protect against inbreeding, leading to the formation of an embryo homozygous for embryonic lethal genes, which subsequently degenerates.
