245 1 0 _aMolecular characterization and similarity relationships among marigold varieties using AFLP markers
490 0 _vSeed Science and Technology, 39(3), p.627-636, 2011
520 3 _aTwenty four marigold varieties selected from two species (12 each)Tagetes erecta L. and Tagetes patula L. were characterized through AFLP markers. Selective amplification of the genomic DNA using 15 polymorphic primer combinations revealed a total number of 506 amplicons of which 468 were polymorphic with an average of 32.07 bands per primer pair. Out of 15 primer pairs three recorded 100 percent polymorphic bands and the remaining produced polymorphic bands in the range of 85.71 to 98.59 percent. The genetic similarity estimates among the varieties ranged between 0.25 to 0.92. The lowest (0.25)similarity coefficient was observed between Sierra yellow and Safari scarlet. The highest (0.92)similarity was recorded between French varieties Safari red and Safari bolero. The cluster diagram based on UPGMA method grouped the marigold varieties into two main clusters species wise. Thus, AFLP analysis showed clear-cut species and varietal differences. confirming the reliability of AFLP analysis and its suitability for the characterization of marigold varieties.
