520 3 _aProtoplasts from pea leaves (Pisum sativum)and vetch cell suspension cultures (Vicia hajastana)were fused by agglutination with polyethylene glycol (PEG 1540). The frequency of heterokaryocytes was a function of the duration of preincubation, i.e. the time lapsed between the removal of cell wall digesting enzymes from the isolated protoplasts and agglutination with PEG. The yield of heterokaryocytes decreased from 9 percent after 5 min to 1 percent after 2 h of preincubation. The most obvious drop occurred between 5 and 15 min of preincubation. The decrease in fusion frequency may be due to rapid cell wall regeneration in vetch protoplasts.
