| 520 | 3 | _aCitrus chlorotic dwarf disease (CCDD) seriously affects the citrus industry. Citrus chlorotic dwarf-associated virus (CCDaV) is speculated to be the causal agent ofCCDD. Howcvcr, this spcculation has not been confirrncd by fulfilling Koch's postulatcs. In this study, an infcctio us clone comprising a 1.6-fold tandem CCDaV genome in the binary vector pBinPLUS was eonstructed and agro-inoculated into 'Eurcka' lemon (Citrus liman) scedlings through vacuum infiltration. At 60 days postinoculation, 25 percent of the 'Eurcka' lcmon seedlings dcvelopcd symptoms of crinkling and curling that wcre the same as those associated with the wild-type virus. Western blotting and graft transmission assays confirrned that the infectious clone systcmically infected 'Eureka' lemon seedlings. In addition, CCDaV can establish infection on thrcc more Citrus species and one hybrid, although at diffcrent infection rates. These findings support that CCDaV is the primary causal agent ofCCDD. The infectious CCDaV clone will allow furthcr studies on the functions ofviral proteins and molecular interactions ofCCDaV with its hosts. | |
