Vacuum liquid chromatographic isolation and characterization of beta-sitosterol from the bark of Sarcocephalus latifolius (Smith Bruce).

The methanol fraction of the stem bark of Sarcocephalus Latifolius (Smith Bruce)was subjected to vacuum liquid chromatography (VLC)isolation method using an isocratic solvent of petroleum ether: ethyl acetate at a ratio of 1:9 by collecting 20ml each and 45 fractions were collected. Based on their Thin Layer Chromatographic (TLC)analysis, fraction 6, 7, 8 and 9 were combined to give SB-C which was further subjected to VLC and eluted isocratically using petroleum ether: ethyl acetate (1:9)to obtain SB-C-1 to SB-C-8 (8 Fraction). Fraction 5 shows a prominent single yellow spot that was cleaned further by Preparative Thin Layer Chromatography (PTLC)to obtain SB-C1. The structure of the compound was established by spectroscopic studies as well as by comparison with existing data. Based on these SB-C1 was proposed to be _-Sitosterol.
