Recursos en línea: Resumen: The introduction of exogenous DNA into yeast by transformation has become an essential technique in molecular biology. Transformation is used to investigate the genomics and proteomics of yeast itself and also when yeast is employed as a system to study the genes and gene products of other organisms. Intact yeast cells can be transformed by a number of procedures: the lithium acetate/single-stranded carrier DNA/polyethylene glycol (LiAc/SS-Carrier DNA/PEG)method, 1-3 elec- troporation, 4-6 agitation with glass beads, 7 and bombardment with DNA-coated microprojectiles. 8 Cells can also be transformed after conversion to spheroplasts by treatment with Zymolyase. 9 We have reviewed the application of these techniques to Saccharomyces cerevisiae and other yeastsJ ° We focus here on the transforma- tion of intact cells by the LiAc/SS-Carrier DNA/PEG method since it is the most widely applicable.
